{"created":"2023-07-25T09:59:05.578824+00:00","id":2330,"links":{},"metadata":{"_buckets":{"deposit":"ac666135-4cd5-41a3-b142-a135d5b71093"},"_deposit":{"created_by":15,"id":"2330","owners":[15],"pid":{"revision_id":0,"type":"depid","value":"2330"},"status":"published"},"_oai":{"id":"oai:oacis.repo.nii.ac.jp:00002330","sets":["187:188:594","187:191:235","70:596","70:597","78:564"]},"author_link":["3631","3626","2252","3628","1460","3633","3627","3629","2860","3625","3632","3630"],"item_10010_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2021-08-27","bibliographicIssueDateType":"Issued"},"bibliographic_titles":[{}]}]},"item_10010_description_25":{"attribute_name":"研究課題番号","attribute_value_mlt":[{"subitem_description":"16K14972","subitem_description_type":"Other"},{"subitem_description":"20H00431","subitem_description_type":"Other"},{"subitem_description":"18K05816","subitem_description_type":"Other"}]},"item_10010_description_26":{"attribute_name":"注記","attribute_value_mlt":[{"subitem_description":"問い合わせ先は、発信当時のものです"}]},"item_10010_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"アユは夏の代表的な川魚であり、多くの河川で友釣りなどが楽しまれている。アユは遊漁用の種苗放流や養殖など、内水面漁業において重要な魚種の一つである。養殖業においては、人気のある子持ち鮎を効率的に作出するために全雌生産(注1)が行われており、その際に遺伝的な性を判別する技術のニーズがある。\nアユはXX-XY型の性決定様式を持ち、雄のY染色体上に存在する性決定遺伝子によって性が決まると考えられる。またアユの属するキュウリウオ目は進化の過程でサケマス類の共通祖先と約1億9千万年前に分岐したと考えられている。さらにキュウリウオ目はメダカやマグロなど多くの魚類が属する真骨魚類の共通祖先と約1億8千万年前に分岐したと考えられている。したがってアユの全ゲノムを調べることで魚類のゲノム進化の過程を知ることができると考えられる。\n本研究ではまずアユの全ゲノムを解読し、他の魚類との比較からその特徴を明らかにした。硬骨魚類の祖先の染色体から融合や分裂を経て、現在のアユの染色体にいたる進化の痕跡を探索した。\nアユゲノムはおよそ4億5千万塩基対とメダカ(8億塩基対)などと比べると小さく、アユではゲノムの縮小化がおこったことが明らかになった。また天然魚を用いた遺伝解析(ゲノムワイド相関解析)により雄特異的なゲノム領域を明らかにした。さらに、その領域に存在する候補遺伝子の発現解析などにより候補遺伝子を絞り込むとともに、ゲノム編集(CRISPR/Cas9法)による遺伝子機能解析を実施した。その結果、Y染色体特異的抗ミュラー管ホルモン型受容体遺伝子がゲノム編集された遺伝的雄個体では、精巣ではなく卵巣が形成され、この遺伝子がアユの性決定遺伝子であることが明らかになった。これにより、アユの遺伝的性判別が正確に実施可能になった。"}]},"item_10010_description_6":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"問合せ先:坂本崇"},{"subitem_description":"プレスリリース掲載論文はこちら:
